Croton lechleri. Family: Euphorbiaceae.

People Use This For:

Orally, sangre de grado, also referrred to by its scientific name croton lechleri, or its constituent, SP-303, is used for treating diarrhea associated with cholera, AIDS, traveling, cancer treatment, Clostridium difficile infection and irritable bowel syndrome (IBS). It is also used orally for supporting the body's tissue repair mechanisms and treating viral respiratory infection. Additionally, sangre de grado is used orally for oropharyngeal and gastrointestinal ulcers, fever, hemorrhage, bleeding gums, fractures, wound healing, hemorrhoids, eczema, insect bites and stings, vaginitis, for vaginal baths before childbirth, and as a general tonic.
Topically, sangre de grado or its constituent, SP-303, is used for treating herpes simplex virus (types 1 and 2).

Safety:

POSSIBLY SAFE ...when SP-303, a derivative of croton lechleri, is used orally or topically and appropriately.

Effectiveness:

POSSIBLY EFFECTIVE
AIDS-related diarrhea. Taking sangre de grado as part of a standardized resin extract containing SP-303 (SB-Normal Stool Formula, ShamanBotanicals.com) orally seems to reduce stool weight and frequency in people with AIDS-related diarrhea.
Herpes simplex virus (HSV). Applying sangre de grado topically as part of a standardized resin extract containing SP-303 seems to be effective for treating genital and anal herpes simplex lesions in people with AIDS. But it doesn't seem to be effective for treating acyclovir-unresponsive mucocutaneous herpes simplex lesions in these patients.
Traveler's diarrhea. Taking sangre de grado as part of a standardized resin extract containing SP-303 seems to be effective for symptomatic treatment of traveler's diarrhea.
INSUFFICIENT RELIABLE EVIDENCE TO RATE
Insect bites and plant reactions. Applying sangre de grado resin alleviated the symptoms of insect bites (fire ants, wasps, bees) and plant reactions in a group of pest control workers. More evidence is needed to rate sangre de grado for this use.

Mechanism of Action:

Sangre de grado is a tree native to the Amazon regions of South America. It is also called sangre de dragon. The resin, a viscous red latex, and bark have a long history of oral and topical medicinal use. Sangre de grado is reported to have anti-inflammatory, antibacterial, anti-hemorrhagic, antiseptic and anti-tumor properties. Topically, it has been shown to reduce vasodilation, swelling and secretory response to irritants, promote mucosal healing, and prevent hyperalgesic responses in the laboratory. Several active constituents of sangre de grado have been isolated, including SP-303 and tapsine. Preliminary evidence suggests that the constituent SP-303 might control diarrhea by inhibiting cyclic adenosine monophosphate(cAMP), which causes chloride and fluid secretion. SP-303 shows activity against types 1 and 2 herpes simplex viruses, respiratory syncytial virus (RSV), and influenza A virus, possibly by inhibiting viral penetration of cells. Tapsine demonstrates anti-inflammatory and wound healing properties when applied topically. Sangre de grado does not stimulate cell proliferation and has not shown carcinogenic activity after 17 months of treatment in experimental models. Preliminary evidence suggests that sangre de grado resin is not cytotoxic, and has antibacterial and pro-oxidant activity.

Adverse Reactions:

Orally, no serious adverse reactions or lab abnormalities have been reported in clinical studies using the sangre de grado derivative SP-303.
Topically, SP-303 can cause local pain and burning.

LEUKEMIA: Preliminary evidence suggests that sangre de grado might prolong survival time of childhood leukemia cells in vitro. Theoretically, taking sangre de grado might worsen disease activity in patients who have leukemia. Advise patients with leukemia not to take sangre de grado.

Dosage/Administration:

ORAL: For treating AIDS-related diarrhea, a sangre de grado extract containing 500 mg of SP-303 (SB-Normal Stool Formula) every 6 hours has been used. For treating traveler's diarrhea, a sangre de grado extract containing 125-500 mg SP-303 (SB-Normal Stool Formula) 4 times daily for 2 days has been used.

Comments:

SP-303 was in Phase III clinical trials with "fast track" designation for treatment of AIDS-related diarrhea and Phase II trials for traveler's diarrhea. After the FDA rejected a new drug application and requested additional clinical testing, the manufacturer, Shaman Pharmaceuticals, became Shaman Botanicals and elected to market SP-303 as a dietary supplement, SB-Normal Stool Formula.
