How Longjack Works?
The applicable parts of long jack or tongkat ali are its root or bark. The most commonly used preparations are extracts and powders. These preparations are known as long jack powder, tongkat ali root extract or tongkat ali powder. The primary active constituents of Eurycoma Longifolia are eurycomanol, longilactone, 11-dehydroklaineanone, dehydrolongilactone, eurycomanone and others. The quassinoids present in Longjack Tongkat Ali are responsible for its bitter taste. Other important constituents of long jack include triterpenes, beta-carboline alkaloids, squalene derivatives, canthine-6-one, and biphenylneolignans.
Quassinoid constituents of Eurycoma Longifolia possess a variety of pharmacological properties like antitumor, antichistosomal and plasmocidal activities. The extracts of Longjack Tongkat Ali have shown to have activity against the strains of malaria, including Plasmodium falciparum, in laboratory. This anti-malarial activity of long jack is attributed to its quassinoids content. Eurycoma Longifolia extracts seem to potential sexual drive, libido and sexual activities in animals. The results of some studies suggest that use of Longjack Tongkat Ali may have an androgenic effect. The use of long jack extracts seems to stimulate the growth of levator ani muscles. Studies on animal models have shown the anxiolytic effects of Eurycoma Longifolia.
Some studies do point towards its potential toxic effects as well. Ethanol extracts are more toxic as compared to water or diethyl ether extracts. Another toxic extract is the butanol extract. Animal studies have shown that eurycomanone constituents in Longjack Tongkat Ali are responsible for its toxicity.

However, the assessment of adverse reactions and safety of Eurycoma Longifolia was not thoroughly assessed in that study. Longjack Tongkat Ali extracts have shown to increase the level of testosterone in the body. Results of both human and animal research has shown that its use might increase the levels of testosterone. If the levels of testosterone were to increase beyond the normal limits, it might cause several testosterone induced adverse effects including hepatotoxicity, resistance to insulin, acne and others.

How Longjack Interacts With Drugs?
Propranolol - This interaction is of moderate nature but still caution should be observed. Results of a clinical trial showed that taking a single dose (200 mg) of a water based extract of Eurycoma Longifolia along with a single dose (80 mg) of Inderal (propranolol) can significantly decrease the level of propranolol. The levels of propranolol were decreased by 29% in that experiment. Moreover, the peak concentration of propranolol was decreased by 42%. The time taken by propranolol to reach its maximum concentration also showed a 86% increase. However, factors like elimination half life of propranolol were not affected. This shows that use of long jack decreases the level of propranolol in the blood by decreasing its absorption from the gut rather than increasing its clearance from the body. However, the way it decreases the absorption of propranolol from the gut is still unknown. It is also unclear if separating the administration would prevent this interaction from happening.
